SON-1410 is a bifunctional recombinant fusion protein developed by Sonnet BioTherapeutics. It consists of an unmodified single-chain human interleukin 18 (IL-18) and an unmodified interleukin 12 (IL-12), linked via the albumin-binding domain of the single-chain antibody fragment A10m3. The construct is designed to target tumor and lymphatic tissue, leveraging the FHAB platform for extended half-life and targeted delivery. Mechanistically, SON-1410 acts as a T lymphocyte stimulant by activating both innate and adaptive immune responses—specifically, it stimulates Th1 cells when combined with IL‑12 and activates natural killer (NK) cells. Preclinical data suggest that synergy between IL‑18 and IL‑12 can upregulate each other's receptors, increase chemokines such as CXCL9/CXCL10, and potentially convert immunologically "cold" tumors into "hot" ones to enhance antitumor immunity. The drug has been investigated primarily for malignant melanoma (preclinical phase) and renal cancer (research phase). SON‑1410 has since been replaced in development by a related molecule, SON‑1411[1][2][3][4][5].
