SON-5010 is a novel antibody-drug conjugate (ADC) under development by Sonnet BioTherapeutics. The construct features an anti-HER2-FHAB-anti-HER2 targeting scaffold (using the company's FHAB albumin-binding single-chain variable fragment technology) linked to a docking peptide, which serves as a conjugation site for three molecules of monomethyl auristatin E (MMAE), a synthetic cytotoxic agent that disrupts the microtubule network and suppresses cell proliferation and mitosis. This non-IgG ADC format is designed to increase tumor accumulation through albumin binding and offers flexibility for different linkers, toxins, and drug-to-antibody ratios (DAR). In preclinical models (e.g., the BT-474 HER2+ breast carcinoma mouse model), SON-5010 showed comparable tumor reduction activity and no detected toxicity relative to approved anti-HER2 ADCs such as Kadcyla and Herceptin-based MMAE ADCs. The platform is designed for versatility, enabling potential bispecific or trispecific tumor targeting capabilities.
