Sonelokimab is a trivalent monomeric nanobody designed to target and neutralize the pro-inflammatory cytokines interleukin 17A (IL-17A), interleukin 17F (IL-17F), and their heterodimer IL-17A/F, which are implicated in the pathogenesis of several autoimmune diseases. It consists of three heavy-chain-only domains—two that bind with high affinity to IL‑17A and IL‑17F, inhibiting the naturally occurring dimers (IL‑17A/A, IL‑17A/F, and IL‑17F/F), and a third domain that binds human albumin to enhance tissue penetration at sites of inflammation[1][3][4][6]. Sonelokimab is being developed primarily for moderate-to-severe hidradenitis suppurativa, psoriatic arthritis, and plaque psoriasis[1][2][3]. Its nanobody structure allows for improved tissue penetration compared to conventional antibodies. The drug is currently under investigation in multiple phase 2 and phase 3 clinical trials[2][7].
