Combination therapy consisting of sonidegib (LDE225), a potent selective and orally bioavailable SMO antagonist that inhibits the Hedgehog signaling pathway, with FOLFIRINOX chemotherapy (fluorouracil, leucovorin, oxaliplatin, and irinotecan). This combination targets both the tumor stroma (via sonidegib) and cancer cells directly (via FOLFIRINOX) in pancreatic cancer. Sonidegib specifically inhibits the Smoothened (SMO) protein, blocking Hedgehog pathway signaling which is involved in the desmoplastic reaction in pancreatic cancer.
