sonkibartinib (also known as SC0191) is a small molecule WEE1 kinase inhibitor. It has demonstrated potent kinase inhibiting activity against WEE1 and significantly inhibited tumor cell proliferation in TP53 mutant NCI-H446 and BxPC-3 cells in preclinical studies. The drug has shown promising antitumor efficacy in preclinical models of TP53 mutant solid cancers, including small cell lung cancer (SCLC) and pancreatic cancer (PC). Developed by Biocity Biopharma, Shijiazhuang Sagacity New Drug Development Co., Ltd., and Wuxi Zhikang Hongyi Biological Technology Co Ltd, sonkibartinib is currently undergoing Phase 1/2 clinical trials for advanced or metastatic solid tumors, with specific indications in metastatic colorectal carcinoma and recurrent ovarian cancer. Its mechanism involves promoting DNA-damaged cells into the M phase, leading to apoptosis in TP53 mutant cancer cells.
