This triplet combination therapy is a chemotherapy-free regimen designed for the treatment of B-cell malignancies, specifically follicular lymphoma. It combines **sonrotoclax** (BGB-11417), a potent and highly selective second-generation BCL-2 inhibitor; **zanubrutinib** (Brukinsa), a next-generation small molecule inhibitor of Bruton's tyrosine kinase (BTK); and **rituximab**, a chimeric monoclonal antibody targeting the CD20 antigen. By simultaneously inhibiting BCL-2-mediated anti-apoptotic pathways, blocking B-cell receptor (BCR) signaling via BTK, and inducing antibody-dependent cellular cytotoxicity (ADCC) through CD20 binding, the regimen aims to achieve synergistic anti-tumor activity. It is currently being evaluated in the Phase II FLOURISH trial for previously untreated follicular lymphoma patients to improve outcomes and avoid the toxicities associated with traditional chemoimmunotherapy.
