sonrotoclax + dexamethasone

**Sonrotoclax + dexamethasone** is an investigational, all-oral combination therapy for patients with relapsed/refractory multiple myeloma harboring a t(11;14) translocation. Sonrotoclax (BGB-11417) is a next-generation, highly selective small molecule inhibitor of BCL2 (B-cell lymphoma 2), promoting apoptosis in cancer cells by blocking BCL2's anti-apoptotic function. Dexamethasone is a corticosteroid commonly used in myeloma regimens for its anti-inflammatory and anti-myeloma activity. In phase 1b/2 studies, this combination has demonstrated a tolerable safety profile with promising efficacy (overall response rates of ~75-80%, very good partial response or better in ~50-56% of patients). The regimen is being investigated in heavily pretreated, t(11;14)-positive relapsed/refractory multiple myeloma.
