Overview

This combination therapy is a therapeutic regimen being evaluated in a Phase II clinical trial (IND245) led by the Canadian Cancer Trials Group (CCTG) for the treatment of relapsed or refractory mantle cell lymphoma (MCL). The regimen consists of sonrotoclax (BGB-11417), a potent and selective next-generation BCL-2 inhibitor, and zanubrutinib (Brukinsa), a next-generation Bruton's tyrosine kinase (BTK) inhibitor, administered in conjunction with standard-of-care chimeric antigen receptor (CAR) T-cell therapy. The combination aims to enhance the efficacy of CAR-T cell therapy by utilizing targeted small molecules to inhibit survival signaling pathways (BCL-2 and BTK) in malignant B-cells, potentially overcoming resistance and improving durable response rates in patients who have failed prior therapies.
