SOP1247 is a small molecule naphthalene diimide derivative that functions as a G-quadruplex (G4) DNA stabilizer. It was developed as part of a medicinal chemistry effort to optimize the G4-binding affinity and anticancer activity of the precursor compound CM03. Structurally, SOP1247 is an analog of the clinical candidate QN-302, distinguished by a methoxy group at the 4th position of the naphthalene diimide core. In preclinical studies, the compound demonstrated nanomolar potency against various cancer cell lines, including gemcitabine-resistant pancreatic ductal adenocarcinoma (PDAC) models such as MIA-PACA2 and PANC-1. Despite its potent G4 stabilizing ability and favorable transcriptional profile, SOP1247 was not advanced to clinical trials, as Qualigen Therapeutics prioritized QN-302 for clinical development due to its superior potency and G4 affinity.
