Overview

This is a combination regimen consisting of three drugs: - **Sorafenib** is an oral small molecule multikinase inhibitor that targets RAF kinases (including BRAF), vascular endothelial growth factor receptors (VEGFR), and platelet-derived growth factor receptor beta (PDGFR-β). It inhibits tumor cell proliferation via the RAF/MEK/ERK pathway and reduces angiogenesis by blocking VEGFR and PDGFR signaling[8]. - **Gemcitabine** is a nucleoside analog used as chemotherapy. It incorporates into DNA during replication, causing chain termination and apoptosis in rapidly dividing cells. - **Interferon** refers to interferons such as interferon-alpha; these are cytokines with immunomodulatory effects that can enhance anti-tumor immune responses. This combination has been explored in clinical trials for advanced cancers such as pancreatic cancer and renal cell carcinoma. The rationale for combining these agents lies in their complementary mechanisms—targeting tumor proliferation pathways (sorafenib), direct cytotoxicity to dividing cells (gemcitabine), and immune modulation (interferon)[2][3]. However, evidence for efficacy remains limited or inconclusive.
