Sorafenib and vorinostat is a combination therapy that demonstrates synergistic anti-cancer effects. Sorafenib is a multi-kinase inhibitor that blocks tumor proliferation by inhibiting the RAF/MEK/ERK pathway and reduces tumor angiogenesis by inhibiting Vascular Endothelial Growth Factor Receptor (VEGFR) and Platelet-Derived Growth Factor Receptor (PDGFR) signaling. Vorinostat (SAHA) is a histone deacetylase inhibitor (HDACI) that promotes cell death through multiple mechanisms. When combined, these drugs interact synergistically to induce cell death in various cancer types, most notably through enhanced activation of the extrinsic apoptotic pathway. The combination works by activating CD95, increasing reactive oxygen species (ROS), calcium, and ceramide levels, which collectively promote apoptosis. This combination has shown efficacy against hepatocellular carcinoma, renal cell carcinoma, pancreatic cancer, and colon cancer cells.
