Drug intelligence / Profile preview
DPH001 is an improved oral formulation of the protein kinase inhibitor sorafenib, developed by Disruptive Pharma using a proprietary mesoporous magnesium carbonate (MMC) drug delivery platform. Sorafenib is approved for the treatment of hepatocellular carcinoma (HCC), renal cell carcinoma (RCC), and certain types of thyroid cancer. The DPH001 formulation leverages MMC to stabilize sorafenib in an amorphous state, enhancing its solubility and bioavailability. Preclinical studies have demonstrated that DPH001 achieves bioequivalent exposure at 50% of the standard active pharmaceutical ingredient (API) dose compared to conventional NEXAVAR, with improved absorption and reduced inter-patient variability. This reformulation aims to provide similar efficacy with potentially fewer side effects due to lower dosing requirements[1][2][5].
