Soralimixin (BRII-693) is a novel synthetic lipopeptide antibiotic being developed by Brii Biosciences for the treatment of critically ill patients with multidrug-resistant (MDR) and extensively drug-resistant (XDR) Gram-negative bacterial infections. As a next-generation intravenous macrocyclic peptide derived from the polymyxin scaffold, it is designed to enhance antibacterial potency against pathogens such as carbapenem-resistant *Acinetobacter baumannii* (CRAB), *Pseudomonas aeruginosa* (CRPA), and *Enterobacterales* (CRE). Its mechanism of action involves targeting and disrupting the bacterial cell membrane. Soralimixin is engineered to provide a superior safety profile compared to older polymyxins by reducing renal and neurotoxicities. It has completed Phase 1 healthy volunteer studies and is progressing toward Phase 3 registrational trials for hospital-acquired and ventilator-associated bacterial pneumonia.
