Overview

Sorfequiline (GSK3657322) is a novel small molecule antimalarial agent currently under development for the treatment of malaria. It functions by inhibiting **Plasmodium falciparum ATPase 4 (PfATP4)**, a P-type sodium-transporting ATPase located on the parasite's plasma membrane. By blocking this pump, sorfequiline disrupts sodium homeostasis within the parasite, leading to a lethal rise in intracellular sodium levels and rapid parasite clearance. Developed by GSK in collaboration with organizations like the Medicines for Malaria Venture (MMV), it is designed as a fast-acting component of next-generation combination therapies to address drug-resistant strains of *Plasmodium falciparum*.
