Overview

SORL1-blocking antibody is an experimental monoclonal antibody that targets Sortilin-related receptor 1 (SORL1), a protein involved in endosomal trafficking and recycling. In ovarian cancer, SORL1 is found to regulate the recycling of integrins (specifically α3/β1) and growth factor receptors (EGFR, FGFR4) to the cell surface, which facilitates cancer cell adhesion, invasion, and mesothelial clearance—key steps in peritoneal dissemination. By inhibiting SORL1, the antibody disrupts these trafficking pathways, leading to the sequestration of integrins in intracellular compartments and reducing the metastatic potential of ovarian cancer cells. This therapeutic strategy is being explored to prevent or treat recurrent ovarian cancer metastasis.
