Overview

Sortilin antibodies are a class of therapeutic monoclonal antibodies designed to bind to sortilin (also known as SORT1 or neurotensin receptor 3), a multi-ligand type-1 receptor that plays a critical role in protein trafficking and lysosomal degradation. In the context of neurodegeneration, sortilin acts as a clearance receptor for progranulin (PGRN); by inhibiting sortilin, these antibodies prevent the degradation of PGRN, thereby increasing its extracellular levels to support neuronal survival and reduce neuroinflammation. This approach is most advanced in treating Frontotemporal Dementia (FTD) caused by GRN mutations, where latozinemab (AL001) is in late-stage clinical trials. Additionally, sortilin antibodies are being explored in oncology, as sortilin is often overexpressed in various cancers and contributes to tumor progression and the internalization of regulatory proteins.
