The sortilin-targeting miniprotein is an affibody-based therapeutic designed to address Frontotemporal Dementia (FTD) caused by progranulin (GRN) mutations. It functions by inhibiting sortilin-mediated degradation of progranulin, thereby increasing extracellular progranulin levels. This miniprotein is presented as a non-immunoglobulin alternative, offering potential advantages such as smaller size and more cost-effective production compared to traditional antibodies. In in vitro functional studies, this affibody-peptide fusion has demonstrated similar potency to latozinemab, another sortilin inhibitor in advanced clinical development for FTD.
