SOT106 is a novel antibody-drug conjugate (ADC) developed by SOTIO Biotech using proprietary antibodies and LigaChem Biosciences' ConjuAll ADC platform technology. It targets leucine-rich repeat containing protein 15 (LRRC15), a cell surface protein overexpressed in various malignant tumors such as sarcoma, osteosarcoma, soft tissue sarcoma, glioblastoma, head and neck squamous cell carcinoma, non-small cell lung cancer, and triple-negative breast cancer. LRRC15 has limited expression in normal tissues but is prevalent in mesenchymal malignancies and other aggressive solid tumors. The payload of the ADC is monomethyl auristatin E (MMAE), a tubulin inhibitor that disrupts microtubule dynamics leading to tumor cell death. Preclinical studies have demonstrated that SOT106 achieves significant tumor regression—including complete responses—in multiple models of LRRC15-positive cancers where first-generation LRRC15-targeting ADCs were ineffective. The drug shows favorable pharmacokinetics, safety profile, stability in vivo, high therapeutic index, and potential for best-in-class efficacy across a broad range of target expression levels[1][2][5][6][7].
