SOT109 is a preclinical antibody-drug conjugate (ADC) being developed by SOTIO Biotech. It targets cadherin-17 (CDH17), a protein highly overexpressed in colorectal cancers and other gastrointestinal cancers (gastric, pancreatic, and esophageal). The ADC utilizes fully human antibodies generated using Biocytogen’s RenMab platform. SOT109 incorporates Synaffix’s proprietary GlycoConnect HydraSpace conjugation technology with the SYNtecan E linker-payload system containing an exatecan payload (DAR=4). This design aims to improve precision targeting, overcome tumor heterogeneity, and enhance efficacy while maintaining tolerability. The exatecan payload is a topoisomerase I inhibitor. SOTIO Biotech plans to enter Phase 1 clinical trials in 2026.
