Sotetinib (SY-5007) is a selective, small-molecule inhibitor of the RET (rearranged during transfection) tyrosine kinase, developed by Shouyao Holdings. It is designed to treat advanced solid tumors harboring RET alterations, such as RET fusions or mutations. These alterations are common drivers in non-small cell lung cancer (NSCLC) and medullary thyroid cancer (MTC). Sotetinib works by binding to the RET kinase domain, thereby blocking downstream signaling pathways that promote tumor cell growth and survival. Clinical data indicates significant anti-tumor activity in RET-altered patients, with a manageable safety profile. It is currently in Phase III development for RET fusion-positive NSCLC in China.
