**Sotiburafusp alfa (HB0025)** is a first-in-class bispecific fusion protein comprising a humanized monoclonal antibody against programmed death-ligand 1 (**PD-L1**) fused to the vascular endothelial growth factor receptor 1 (**VEGFR1**) extracellular domain, acting as a **VEGF trap**. Developed by **Huabo Biopharm Co Ltd** (a subsidiary of Zhejiang Huahai Pharma), it simultaneously blocks **PD-L1** to reinvigorate T-cell anti-tumor immunity and sequesters **VEGF** to inhibit angiogenesis, demonstrating superior preclinical tumor growth inhibition compared to separate anti-PD-L1 mAb or VEGFR1D2 fusion protein. Currently in **Phase 2** trials in China for advanced solid tumors including **triple-negative breast cancer**, **squamous non-small cell lung cancer**, **advanced endometrial carcinoma**, **renal cell carcinoma**, and others, with promising efficacy and safety in combination with chemotherapy; Phase 3 trials planned for 2025 in NSCLC and endometrial cancer.[1][4][10][13]
