Overview

Sotigalimab (APX005M) is a novel humanized IgG1κ monoclonal antibody that acts as an agonist of the CD40 receptor. It binds with high affinity to human and monkey CD40 but not to mouse or rat CD40. Sotigalimab blocks the binding of CD40 ligand (CD40L) to its receptor by targeting specific epitopes on the CD40 protein. This blockade leads to potent activation of antigen-presenting cells (APCs), such as dendritic cells, monocytes, and B-cells. The activation results in upregulation of co-stimulatory molecules and cytokine release from immune cells, thereby enhancing T cell proliferation and stimulating both innate and adaptive anti-tumor immune responses. Sotigalimab is being developed primarily for oncology indications including pancreatic cancer, esophageal cancer, gastroesophageal junction cancer, melanoma, non-small cell lung cancer (NSCLC), rectal cancer, sarcoma—often in combination with other immunotherapies or chemotherapies[1][5][6][9].
