Overview

This is a triple combination therapy consisting of sotigalimab (APX005M), cabiralizumab, and nivolumab. Sotigalimab is a CD40 agonist antibody that binds with high affinity to the CD40 ligand-binding site on CD40 and is optimized for selective interaction with Fcγ receptors to enhance agonistic potency. It acts as a master regulator of immunity by mobilizing multiple arms of the immune system to initiate CD8+ T-cell-mediated responses against tumors. Cabiralizumab is a CSF1R inhibitor, while nivolumab is a PD-1 inhibitor that blocks the PD-1 pathway to help prevent cancer cells from hiding from T cells. This combination was investigated for patients with anti-PD-1/PD-L1-resistant tumors, particularly in melanoma, non-small cell lung cancer (NSCLC), and renal cell carcinoma (RCC).
