Sotorasib is a first-in-class, orally available small molecule inhibitor that selectively and irreversibly targets the KRAS G12C mutation, a specific alteration in the KRAS gene found in certain cancers. By covalently binding to the cysteine residue at position 12 of mutant KRAS (G12C), sotorasib locks the protein in its inactive GDP-bound state, thereby inhibiting downstream oncogenic signaling pathways. This leads to reduced tumor cell proliferation and survival. Sotorasib is primarily indicated for adults with locally advanced or metastatic non-small cell lung cancer (NSCLC) or metastatic colorectal cancer harboring a KRAS G12C mutation who have received prior systemic therapy. It is also being studied for other cancers with this mutation. The drug was developed by Amgen.
