**Sotorasib + AMG 404** is an investigational combination therapy for cancer treatment, currently studied in advanced solid tumors (especially non-small cell lung cancer with KRAS G12C mutation). Sotorasib is a small molecule that selectively and irreversibly inhibits the KRAS G12C mutant protein, blocking KRAS-dependent signaling pathways and thereby inhibiting tumor growth. AMG 404 is a fully human immunoglobulin G1 (IgG1) monoclonal antibody directed against the programmed cell death 1 (PD-1) receptor; it blocks the interaction between PD-1 and its ligands PD-L1 and PD-L2, thereby promoting immune-mediated anti-tumor activity. The combination seeks to maximize anti-tumor efficacy by simultaneously targeting oncogenic signaling (KRAS G12C) and enhancing immune response via checkpoint inhibition. Both agents are developed by Amgen[2][3][5].
