Overview

Sotuletinib (BLZ-945) is an orally active, potent and selective small molecule inhibitor of the colony-stimulating factor 1 receptor (CSF-1R). It binds to CSF-1R expressed on tumor-associated macrophages (TAMs), blocking its activity and downstream signal transduction pathways. This inhibition reduces the proliferation and survival of TAMs and reprograms their immunosuppressive phenotype. Sotuletinib has demonstrated brain penetration and is being investigated for various indications including glioma, amyotrophic lateral sclerosis (ALS), multiple sclerosis, brain cancer, and as a potential adjunct in HIV eradication strategies targeting viral reservoirs in the brain. The drug was developed by Novartis[1][2][3][5][6].
