Overview

Sovesudil (AMA0076, PHP-201) is a potent, locally acting small molecule inhibitor of Rho-associated protein kinase 1 (ROCK1) and ROCK2, developed for the treatment of glaucoma and ocular hypertension. It is specifically engineered as a "soft drug," meaning it is active at the site of administration in the eye but is rapidly hydrolyzed by endogenous esterases into an inactive metabolite (AMA0078) upon leaving the target tissue. This design is intended to maximize therapeutic efficacy in lowering intraocular pressure (IOP) by increasing aqueous humor outflow through the trabecular meshwork while significantly reducing common side effects associated with ROCK inhibitors, such as conjunctival hyperemia (eye redness). Originally developed by Amakem NV and later progressed by PH Pharma, sovesudil has demonstrated significant IOP-lowering potential in clinical trials and has advanced into Phase 3 development for primary open-angle glaucoma and related conditions.
