Overview

Sovilnesib is an orally bioavailable, small-molecule inhibitor of the human kinesin-like protein KIF18A, a mitotic kinesin-8 motor protein involved in chromosome positioning during cell division. By selectively inhibiting KIF18A’s microtubule-stimulated ATPase activity, sovilnesib disrupts normal mitotic processes, leading to multipolar cell division, cell cycle arrest, and apoptosis in cancer cells. This mechanism is particularly relevant for cancers characterized by chromosomal instability (CIN), such as high-grade serous ovarian cancer (HGSOC), where tumor cells are highly dependent on KIF18A for proliferation and spindle integrity. Sovilnesib was originally developed by Amgen and is now being developed by Volastra Therapeutics under license from Amgen; BeiGene has also been listed as a developer. The drug has received FDA Fast Track designation for ovarian cancer and is currently in phase I clinical trials for ovarian cancer and other solid tumors[1][2][3][4][5][6].
