SOX + paclitaxel + PD-1 antibody is a combination regimen used in oncology, particularly for the treatment of advanced or metastatic gastric cancer. The regimen consists of three components: - SOX, which combines S-1 (an oral fluoropyrimidine derivative) and oxaliplatin (a platinum-based chemotherapeutic agent), - Paclitaxel, a taxane-class microtubule inhibitor, - A PD-1 antibody (such as nivolumab, pembrolizumab, camrelizumab, or others), which is an immune checkpoint inhibitor targeting the programmed cell death protein 1 pathway. The mechanism of action involves: - S-1 inhibiting thymidylate synthase to disrupt DNA synthesis, - Oxaliplatin causing DNA crosslinking and apoptosis, - Paclitaxel stabilizing microtubules to prevent cell division and induce apoptosis[6][8][10], - The PD-1 antibody blocking the interaction between PD‑1 on T cells and its ligands on tumor cells to enhance anti-tumor immune responses[7][9]. This combination aims to maximize cytotoxic effects against tumor cells while also stimulating the immune system’s ability to recognize and destroy cancer cells. It has been investigated in clinical trials for improved efficacy over chemotherapy alone in gastric cancer[3][5].
