Drug intelligence / Profile preview
Sox9 mRNA-LNP is an investigational lipid nanoparticle (LNP) formulation encapsulating messenger RNA (mRNA) that encodes SRY-box transcription factor 9 (SOX9), a master transcription factor essential for chondrogenesis and cartilage homeostasis. Developed by researchers at institutions including Kyungpook National University, National Taiwan University, and Peking Union Medical College Hospital, Sox9 mRNA-LNP is being evaluated for the treatment of osteoarthritis (OA) and cartilage regeneration. By delivering Sox9 mRNA directly into the joint via intra-articular injection, the therapy aims to restore chondrocyte homeostasis, promote extracellular matrix (ECM) synthesis (such as type II collagen and aggrecan), and suppress pathological Wnt signaling and catabolic enzymes like MMP13. Preclinical studies in rat models of osteoarthritis have demonstrated that intra-articular administration of Sox9 mRNA-LNP significantly attenuates pain behavior and preserves articular cartilage structure.
