Overview

Sozinibercept is a novel recombinant fusion protein developed by Opthea for the treatment of retinal vascular diseases, primarily neovascular (wet) age-related macular degeneration (AMD) and diabetic macular edema (DME). It acts as a "trap" inhibitor targeting vascular endothelial growth factors C and D (VEGF-C and VEGF-D), which are key mediators of angiogenesis and vascular leakage in the retina. Sozinibercept is a soluble form of VEGFR-3 fused to an immunoglobulin G1 Fc fragment, designed to bind and neutralize VEGF-C and VEGF-D, thereby inhibiting their activity on endogenous receptors VEGFR-2 and VEGFR-3. This mechanism complements standard anti–VEGF-A therapies by providing broader inhibition across the VEGF family. Sozinibercept is administered via intravitreal injection, typically in combination with anti–VEGF-A agents such as ranibizumab or aflibercept. Clinical trials have shown that this combination can lead to superior visual outcomes compared to anti–VEGF-A monotherapy[1][2][5][6][7].
