SP-009

SP-009 is a small molecule oral softgel capsule developed by Sunny Pharmtech for the treatment of Transthyretin Amyloid Cardiomyopathy (ATTR-CM). It is currently in the bioequivalence (BE) study phase, which is a stage in generic drug development typically corresponding to Phase 1 clinical trials. SP-009 is designed as a generic version of tafamidis (specifically the free acid form marketed as Vyndamax). Its mechanism of action involves binding to the thyroxine-binding sites of the transthyretin (TTR) tetramer with high affinity and selectivity. This binding stabilizes the TTR tetramer and prevents its dissociation into monomers, which is the rate-limiting step in the pathogenesis of TTR amyloidosis. By preventing tetramer dissociation, SP-009 inhibits the formation of amyloid fibrils that deposit in the heart, thereby slowing the progression of cardiomyopathy.
