Drug intelligence / Profile preview
SP-1072 is a synthetic 16-amino-acid peptide (sequence: YKSTVIVNNKEKTTAE-PEG4-K(N3)-CONH2) that mimics the trans-homophilic interaction motif of domain 1 of CD31 (platelet endothelial cell adhesion molecule-1, PECAM-1). Developed by AlchiMedics in collaboration with INSERM, Université Paris Cité, Université Sorbonne Paris Nord, and the University of Galway, SP-1072 is designed as a biomimetic coating for blood-contacting medical implants, such as cobalt-chromium stents and nitinol flow diverters. By mimicking the surface of a healthy endothelium, the peptide engages intact CD31 molecules on endothelial cells, platelets, and leukocytes, delivering a 'leave-me-alone' signal that prevents platelet adhesion, thrombosis, and local inflammation. This drug-free, passive surface modification promotes rapid, non-inflammatory endothelialization and vascular healing, potentially allowing for the safe reduction or de-escalation of dual antiplatelet therapy (DAPT) in patients undergoing percutaneous coronary intervention (PCI), particularly those at high bleeding risk.
