Drug intelligence / Profile preview
SP-1353 is an orally available, potent, and selective small molecule agonist of the parathyroid hormone 1 receptor (PTH1R). Developed by Septerna, the compound is designed to mimic the action of parathyroid hormone (PTH) to regulate calcium and phosphate homeostasis. In preclinical models of hypoparathyroidism, such as thyroparathyroidectomized (TPTx) rats, oral administration of SP-1353 demonstrated sustained control of serum calcium and phosphate levels within the therapeutic range, as well as modulation of key bone turnover biomarkers. SP-1353 represents a potential oral alternative to injectable PTH peptide therapies for the treatment of hypoparathyroidism and other PTH-related disorders.
