Overview

SP-3164 is an orally administered, next-generation molecular glue and targeted protein degrader developed as a deuterium-stabilized (S)-enantiomer of avadomide (CC-122). It acts by binding to cereblon (CRBN), a component of the CRL4 E3 ubiquitin ligase complex, thereby inducing the recruitment and subsequent degradation of hematological transcription factors Ikaros (IKZF1) and Aiolos (IKZF3). This mechanism leads to selective elimination of cancer-promoting proteins, resulting in apoptosis and reduced viability in cancer cells. Preclinical studies have shown that SP-3164 demonstrates superior antitumor activity compared to other approved molecular glues such as lenalidomide and pomalidomide, both as a single agent and in combination with dexamethasone. The drug is being developed primarily for hematologic malignancies including non-Hodgkin's lymphoma and multiple myeloma[1][3][5][6][7].
