Overview

SP-3165 is a small molecule and the deuterium-stabilized (R)-enantiomer of avadomide (AVA). Developed by Salarius Pharmaceuticals, it serves as the inactive counterpart to SP-3164, the (S)-enantiomer. In preclinical studies for multiple myeloma, SP-3165 has been shown to lack the ability to bind Cereblon (CRBN) or degrade the transcription factors Ikaros and Aiolos. Consequently, it does not induce apoptosis in cancer cells and has demonstrated minimal tumor growth inhibition in mouse xenograft models, where it was observed to potentially support tumor growth instead. It is primarily used as a negative control to demonstrate the stereospecific activity of the active (S)-enantiomer in the molecular glue degrader program.
