SP1049C

**SP1049C** is an investigational polymeric micelle formulation of the anthracycline **doxorubicin**, developed by Supratek Pharma to address multidrug-resistant cancers. Doxorubicin is noncovalently incorporated into mixed micelles formed from the nonionic poloxamers Pluronic L61 and Pluronic F127. The formulation retains doxorubicin's DNA-intercalating and DNA topoisomerase II-inhibitory cytotoxic activity while the Pluronic excipients increase intracellular drug accumulation and impair ATP-dependent P-glycoprotein drug efflux. SP1049C completed Phase 1 and Phase 2 clinical studies in advanced adenocarcinoma of the esophagus and gastroesophageal junction, and received FDA orphan-drug designation for gastric cancer.
