SP142 is a recombinant rabbit monoclonal antibody clone used in immunohistochemistry (IHC) to detect the expression of Programmed Death-Ligand 1 (PD-L1) in tumor cells and tumor-infiltrating immune cells. Developed by Spring Bioscience and marketed by Ventana Medical Systems (a Roche company), it is an FDA-approved companion diagnostic (CDx) specifically validated for use with the immunotherapy atezolizumab (Tecentriq). The assay is used to identify patients with triple-negative breast cancer (TNBC), urothelial carcinoma, and non-small cell lung cancer (NSCLC) who are most likely to benefit from PD-L1/PD-1 checkpoint inhibitor therapy. Unlike other PD-L1 IHC clones (such as 22C3 or 28-8), SP142 is known for its specific staining pattern on immune cells, which is a critical component of the scoring algorithm for certain indications.
