Drug intelligence / Profile preview
SPARC0921 is a gastro-retentive formulation of baclofen developed by Sun Pharma Advanced Research Company (SPARC) for the treatment of spasticity, particularly in patients with multiple sclerosis or spinal cord injury. It utilizes a proprietary Gastro-Retentive System (GRS) technology designed to extend the residence time of the drug in the stomach, allowing for controlled release into the upper gastrointestinal tract where baclofen is most efficiently absorbed. This formulation aims to provide more stable plasma concentrations and potentially reduce the dosing frequency compared to immediate-release baclofen, which typically requires multiple daily doses. Baclofen acts as an agonist at GABA-B receptors, leading to hyperpolarization of motor neurons and inhibition of polysynaptic and monosynaptic reflexes at the spinal level.
