Canakinumab is a recombinant human monoclonal antibody that specifically binds interleukin-1β and neutralizes its inflammatory activity, indicated for inflammatory disorders and also explored in cancer immunotherapy settings[2][4][6]. This combination is under investigation for advanced solid tumors, particularly in cancers resistant to immune checkpoint monotherapy; the rationale is to target multiple immune pathways (PD-1 and LAG-3) and modulate the tumor microenvironment (IL-1β inhibition)[1][5].
