Overview

This is an investigational combination of three humanized monoclonal antibodies for cancer immunotherapy. **Spartalizumab (PDR001)** is an IgG4 humanized monoclonal antibody that binds to PD-1, blocking its interaction with PD-L1 and PD-L2, thus restoring antitumor T-cell activity. **LAG525** is an IgG4 humanized monoclonal antibody that targets LAG-3, inhibiting its interaction with MHC class II, restoring effector T cells and synergizing with PD-1 blockade. **MCS110 (lacnotuzumab)** targets and neutralizes colony-stimulating factor 1 (CSF-1), which is involved in macrophage-mediated resistance to immunotherapies, potentially reducing tumor-associated macrophages and improving the efficacy of checkpoint inhibitors. This combination was studied in advanced/metastatic triple-negative breast cancer and possibly other solid tumors, but development of MCS110 was terminated before full evaluation[5][8].
