Overview

SPC-14 is a novel investigational drug in preclinical development for the treatment of Alzheimer’s disease and related dementia. It is an intranasal formulation that combines ketamine with an FDA-approved therapeutic, designed to target cognitive and neuropsychiatric symptoms associated with Alzheimer’s. The mechanism of action involves dual modulation of the glutamate receptor NDMAR (NMDA receptor) and serotonin type 4 receptor (5-HT4). Preclinical studies have shown that SPC-14 can improve memory function, reduce anxiety-related behaviors, and attenuate stress responses in animal models. The drug has demonstrated efficacy against luteinizing hormone (LH)-induced behavioral changes such as learned helplessness, perseverative behavior, and hyponeophagia. Developed under a sponsored research agreement with Columbia University and exclusively licensed to Silo Pharma, SPC-14 aims to address significant unmet needs in neurodegenerative disease therapy by leveraging both traditional therapeutics and psychedelic research[1][3][4][5][6][7][8].
