The SPCN neoantigen vaccine is a personalized cancer immunotherapy platform developed by researchers at the University of Hong Kong. It utilizes a self-assembling peptide conjugates nanoparticle (SPCN) system to deliver tumor neoantigens identified via artificial neural network (ANN) prediction. These neoantigen peptides are engineered by conjugation with a Toll-like receptor 7 agonist (TLR7a) adjuvant, which facilitates the activation of antigen-presenting cells (APCs). The platform is designed to overcome common limitations of synthetic peptide vaccines, such as low immunogenicity and short half-life, by providing protection and sustained release of the antigens within the body. Preclinical studies in mouse models of melanoma, bladder cancer, and liver cancer have demonstrated that the SPCN platform enhances APC activation in draining lymph nodes and stimulates robust systemic anti-tumor immune responses, characterized by increased interferon-gamma production.
