sPD1-p24fc is a DNA vaccine encoding a fusion protein that links the soluble extracellular domain of Programmed Death-1 (sPD1) to the HIV-1 GAG p24 antigen and an Fc fragment, designed for in vivo targeting of dendritic cells (DCs) via PD-L1/PD-L2. This vaccine aims to amplify adaptive immune responses by cross-presenting the HIV-1 p24 antigen to CD8+ T cells and inducing potent, durable, polyfunctional, cytotoxic, and protective GAG-specific CD8+ T cell immunity. Its mechanism involves DC targeting to enhance vaccine efficacy, particularly important for HIV-1 infection, as p24-specific CD8+ T cells play a role in viral containment. The vaccine is delivered by electroporation of the DNA construct and has been described as uniquely immunogenic, with minimal adjuvant effect from the Fc fusion and critical dependence on the sPD1-p24 structure for immune enhancement[1][3][5][7][9].
