Drug intelligence / Profile preview
SPEEDesign RBD vaccine refers to a family of SARS-CoV-2 vaccine candidates designed using the SPEEDesign (Stabilizer for Protein Expression and Epitope Design) computational platform. Developed by the National Institute of Allergy and Infectious Diseases (NIAID), this approach introduces stabilizing amino acid mutations into the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein. These mutations enhance neutralizing antibody titers, focus the immune response toward structured neutralizing epitopes, increase production yields, and improve antigen stability. A key candidate, noNAG-RBD-E2p, is a stabilized, non-glycosylated RBD genetically fused to a 60-copy self-assembling dihydrolipoyl acetyltransferase (E2p) nanoparticle platform. In preclinical studies, this nanoparticle vaccine elicited potent neutralizing antibody responses in mice and non-human primates, showing robust activity against emerging variants such as Omicron BA.5.
