Overview

Spermine NONOate is a diazeniumdiolate nitric oxide (NO) donor. Under physiological conditions, it spontaneously dissociates to release approximately two moles of NO per mole of the parent compound in a pH- and temperature-dependent manner, with a half-life of approximately 39 minutes at 37°C (pH 7.4). Spermine NONOate is widely utilized as a pharmacological research tool to investigate the physiological and pathophysiological roles of nitric oxide, including vasodilation, angiogenesis, biofilm dispersal, and cellular signaling. It acts primarily by activating soluble guanylate cyclase (sGC), which increases intracellular cyclic GMP (cGMP) levels, leading to smooth muscle relaxation and other downstream effects. It has also been studied for its ability to inhibit O6-methylguanine-DNA methyltransferase (MGMT) via nitrosylation, sensitizing cancer cells to alkylating agents.
