Overview

SPG-178 is a neutral, synthetic self-assembling peptide (SAP) hydrogel developed as a versatile scaffold for tissue engineering and regenerative medicine. It consists of short peptides that spontaneously form a stable, transparent nanofiber hydrogel in a neutral pH environment upon exposure to electrolytes. Unlike earlier SAP hydrogels such as RADA16, which are acidic, SPG-178 maintains a neutral pH, significantly reducing tissue irritation and inflammation. The material is non-biological, non-cytotoxic, and biocompatible, making it suitable for a wide range of clinical applications. It has been investigated as a topical hemostatic agent, a vitreous substitute in ophthalmic surgery, a scaffold for bone and periodontal regeneration, a neuroprotective agent in spinal cord injury, and a tool to prevent pancreatic fistulas after surgery. Its mechanical properties, including viscoelasticity, can be tuned by adjusting peptide and salt concentrations to suit specific biomedical needs.
