Overview

SPH7854 is a potent and selective small molecule antagonist of the Retinoic Acid Related Orphan Nuclear Receptor gamma t (RORγt), developed as a gut-limited therapy for inflammatory bowel disease (IBD). By antagonizing RORγt, SPH7854 inhibits the differentiation of Th17 cells and reduces interleukin-17A (IL-17A) secretion, while promoting regulatory T cell (Treg) differentiation. This mechanism leads to reduced inflammation in preclinical models of colitis. Pharmacokinetic studies indicate that SPH7854 has minimal systemic exposure after oral administration, suggesting improved safety by limiting off-target effects such as thymic lymphoma or liver dysfunction associated with systemic RORγt inhibition. The drug is currently in phase 1 clinical trials for IBD and healthy volunteers[1][3][4][7].
