Drug intelligence / Profile preview
Sphingosine-1-phosphate-enriched high-density lipoprotein (S1P-enriched HDL or S1P-loaded HDL) is an experimental therapeutic preparation of high-density lipoprotein (HDL) loaded with sphingosine-1-phosphate (S1P). In patients with coronary artery disease (CAD) or type 2 diabetes, HDL particles often exhibit a depleted S1P content, which impairs their normal cardioprotective and vasodilatory functions. By loading these dysfunctional HDL particles with S1P, their lipoprotective and cardioprotective properties can be restored. This therapeutic approach has been investigated in preclinical models of acute myocardial infarction (AMI), where S1P-enriched HDL significantly reduced infarct size and improved cardiac ejection fraction. The mechanism of action is mediated through the activation of S1P receptors (such as S1PR1 and S1PR3) and requires the HDL receptor scavenger receptor class B member 1 (SR-BI).
