Overview

SphK1-shRNA is a short hairpin RNA (shRNA) developed for gene therapy research to specifically silence the expression of sphingosine kinase 1 (SphK1), an enzyme implicated in cancer cell proliferation, survival, and metastasis. By downregulating SphK1, SphK1-shRNA inhibits key signaling pathways such as the STAT3 pathway, resulting in decreased tumor cell proliferation, increased apoptosis, and impaired migration and invasion, particularly demonstrated in neuroblastoma and thyroid cancer models. SphK1-shRNA is commonly delivered using lentiviral vectors in preclinical studies, offering a targeted approach to disrupt sphingolipid metabolism and downstream oncogenic signaling[2][4].
